Mutation Surveyor (MS) is a mutation (or SNP) detection software developed under the corporation of both the Softgenetics Inc. (in US) and TodaySoft Inc. in order to discover the DNA mutation (or SNP) with high throughput, high sensitivity and high accuracy. At present, the software is being used by hundreds of well-known organizations all over the world (Famous foreign experts' opinions on MS). Five articles have published respectively in Sciences and Nature reporting the achievements of studies on the DNA mutations with MS (or Mutation Explorer) in less than one year.

MS adopts two original technologies of the world in the field of mutation detection:

a)
|
Anti-correlation technology: By using anti-correlation technology, MS directly discovering the mutations through DNA trace files in contrast to other traditional software's reliance on DNA "base calls". MS compares the DNA traces of the sample sequences with the reference sequences in a point-to-point way and produces Anti-correlation curve. When comparing sample DNA sequences and standard reference sequences, MS uses five independent parameters as the criteria for the determination of mutations. Anti-correlation technology can effectively eliminate the interference of background noise and accurately analyze DNA traces produced by different DNA sequencers.

b)
|
Correct identification of heterozygous indels: The analysis and judgment of heterozygous indels is a challenge for all types of mutation detection software. MS successfully resolves this global problem.
|
|
|
In a heterozygous indels sample, maybe the 5' terminal of sequence is quite good, however, in the middle part, the peaks suddenly turns disorderly, and the base calling results may contains a lot of error information or N. Most of the mutation analysis software will produce several false positive results when analyzing those samples. But on the analysis of MS, the disorderly peaks will show their true features by anti-correlation processing with the software; wild-type sequences and indels-mutant sequences will be indicated in turn; the deviation of mutant sequences resulting from insertion or deletion can be observed obviously. Through comparing the two types of sequences we can clearly see the position of mutant.

High sensitivity
|
|
|
|
For bi-directional sequenced samples, MS's analysis has very high rate of accuracy and sensitivity (>99.9%). For unidirectional sequenced samples, it has also a rate of more than 96%. The software can detect the mutation easily if heterozygous peaks are higher than the main peaks by 5%. The user can easily obtain the mutation position, bases and other various parameters from the anti-correlation curve.

According to the consistency of DNA duplex mutations, gene variations may be divided into homozygous variation and heterozygous variation; and the types of variation may be divided into insertion, deletion and substitution, which can be seen from the DNA sequence charts(chart 1,2,3,4)

High-throughput analysis
|
|
|
|
MS is suitable for not only the analysis of the result of small sample sequencing but also high-throughput analysis. The software can analyze 400 or more sequences at one time, supporting the preset groups and can carry out automatic grouping for processing sequences of any amount. You only need to conduct simple setting according to the software's prompts and then leave the other work to the software for automatic processing.

|
MS reports the result of analysis in three concise formats:

1. According to forward and reverse sequence pairs
|
|
2. According to the global analysis of all samples
|
|
3. One-sample analysis
|
|
|
|
Each format is linked to and displayed in electropherograms for rapidly confirming the result of discovery.
|
|
|
|
Reports are linked with electropherograms, which, from top to bottom, comprise forward reference curve, forward sample curve, forward mutation curve, reverse reference curve, reverse sample curve and reverse mutation curve. Mutations are clearly shown in the forward and reverse curves in the form of spikes.

The user may choose to print the samples used or only the mutation samples .

All data can be easily output to LIMS system or electronic datasheet.
